Understanding and engineering peptide–membrane interactions
Membrane-active peptides interact with lipid bilayers through a combination of electrostatic attraction, hydrophobic interactions, peptide conformation and lipid organisation. These interactions can influence membrane binding, insertion, disruption, permeability and selectivity.
At Peptide Intelligence Consulting, we help researchers understand the molecular factors governing peptide–membrane interactions and develop informed strategies for tuning membrane activity.
The scientific challenge
A peptide that interacts strongly with a membrane does not necessarily interact selectively or safely. Small changes in sequence, charge distribution, hydrophobicity or conformation may alter membrane association and the balance between activity and cellular compatibility.
The challenge is to understand which molecular features support the intended function and which may contribute to unwanted interactions.
Peptide–membrane interaction analysis
Assess how charge and hydrophobicity may influence membrane association.
Examine the role of amphipathicity and conformational preferences.
Consider how lipid composition may influence peptide interactions.
Membrane activity and selectivity
Develop hypotheses about membrane binding, insertion and disruption.
Explore design variables that may influence selectivity between different membrane environments.
Identify potential trade-offs between membrane activity and cellular compatibility.
Environment-responsive peptide design
Explore pH-responsive changes in peptide charge or conformation.
Assess design concepts for peptides intended to change activity in different environments.
Investigate principles relevant to endosomal membrane interactions and intracellular delivery.
Experimental and computational strategy
Recommend suitable approaches for studying peptide–membrane interactions.
Help interpret relevant data from membrane-binding, leakage, imaging or biophysical studies.
Identify complementary experimental and modelling methods.
We connect peptide sequence features with the properties of the target membrane and the intended biological or materials function. We then identify testable hypotheses and recommend appropriate methods for evaluating them.
Depending on the project, this may involve physicochemical analysis, peptide characterization, lipid model systems, biophysical measurements or molecular simulation.
Computational predictions and model membrane results are treated as evidence to guide experiments, not as substitutes for biological validation.
